Identification of age-related resistance loci in cucumber by Quantitative Trait Locus sequencing (QTLseq)
QTLseq is a relatively new, modified version of Bulk Segregant Analysis (BSA) for use with next generation sequencing read data. Using this analysis, we expect to identify QTL associated with ARR to P. capsici in cucumber. We will use these data alongside our previously described RNAseq and metabolomics data to identify genes of interest contributing to ARR to P. capsici. To this end, we are screening a Doubled Haploid population derived from crosses of lines that do and do not express ARR. Screening of detached fruit is performed in plastic trays lined with wet paper towels. Fruit are then inoculated with P. capsici zoospore suspension and trays are covered and sealed. After 7 days of incubation we photograph and rank the disease severity. Bulks of resistant and susceptible lines will be selected and sequenced. QTLseq analysis will be performed using the "QTLseqr" R package which I developed.
